Abstract: O câncer de mama apresenta alta incidência entre mulheres e permanece como um dos principais desafios da saúde pública em todo o mundo. A Galectina-3, uma lectina multifuncional com afinidade por β-galactosídeos, atua em tumores mamários devido à sua capacidade de promover interações célula-célula e célula-matriz extracelular. Portanto, o objetivo deste estudo foi investigar o impacto do silenciamento da Galectina-3 em células de carcinoma mamário 4T1, avaliando seu secretoma em comparação ao controle. Os resultados indicam que o silenciamento promoveu aumento da migração celular e alterações em moléculas de adesão como no versican, sugerindo papel modulador na biologia tumoral. As proteínas com abundância reduzida na linhagem silenciada apresentaram associação ao metabolismo de hexoses e glicose, com forte interação com Eno1, AldoA, AldoC e GAPDH, enzimas-chave da glicólise e gliconeogênese. No entanto, proteínas como MMP3, MMP10, THBS1 e DAG-1 mostraram maior abundância, indicando uma possível implicação na remodelação da matriz extracelular e potencial influência na metástase. Os achados demonstram, pela primeira vez, que o silenciamento de Galectina-3 altera significativamente o secretoma de células 4T1, favorecendo características metastáticas, como evidenciado por ensaios in vivo e in vitro.
Abstract: Breast cancer has a high incidence among women and remains one of the major global public health challenges. Galectin-3, a multifunctional lectin with affinity for β-galactosides, plays a role in breast tumors due to its ability to mediate cell–cell and cell–extracellular matrix interactions. Therefore, this study aimed to investigate the impact of Galectin-3 silencing in 4T1 murine breast carcinoma cells by analyzing their secretome in comparison to control cells. The results indicate that Galectin-3 silencing led to increased cell migration and changes in adhesion molecules such as versican, suggesting a modulatory role in tumor biology. Proteins with reduced expression in the silenced lineage were associated with hexose and glucose metabolism, showing strong interactions with Eno1, AldoA, AldoC, and GAPDH, which are key enzymes in glycolysis and gluconeogenesis. In contrast, proteins such as MMP3, MMP10, THBS1, and DAG-1 were upregulated, indicating potential involvement in extracellular matrix remodeling and metastatic progression. These findings demonstrate, for the first time, that Galectin-3 silencing significantly alters the secretome of 4T1 cells, promoting metastatic features, as evidenced by both in vitro and in vivo assays.
