Resumo

The use of polymer particles that can be recognized by cellular receptors can ensure the success of in situ drug delivery applications. Thus, the objective of this work is to develop functionalized polymer nanoparticles capable of permeating blood-brain barrier cells for drug delivery, aiming at the treatment of neurodegenerative diseases. The technique of copolymerization in miniemulsion allowed the production of nanometric particles from the monomers methyl methacrylate, acrylic acid and methacrylic acid for later functionalization, with average size around 60 to 100 nm. In addition, the encapsulation of clioquinol with an efficiency greater than 90% was proven. The methodology of immobilization by chemical reaction, with 1-ethyl-3- (3-dimethylamino propyl) carbodiimide, was able to promote the functionalization of the particles with the insertion of the peptide trans-activating transcriptor. The produced material was evaluated in terms of the ability to transpose the brain barrier by means of in vitro and in vivo tests. The results showed that the particles produced can penetrate the blood-brain barrier successfully, having been found in the brain tissue of mice. Due to the nonselective nature of the model peptide used, the nanoparticles also cross other tissues, being found in other organs. The half-life in the animals is small, being removed from the circulation after 30 minutes.
